From the unexpected incident in Donatello's laboratory, everything changed for Leonardo. The unknown substance with which his brother had been experiencing had made him a female mutant turtle. The physical changes were notable: his plastron showed some breasts, his shell seemed more sensitive than usual, and his hips, although narrow, had accentuated them, giving him a female figure. His thinner waist gave him surprising agility and flexibility, something he could take advantage of in combat, although he still did not get used to his new reality.

The most uncomfortable for "Leonarda" - as Michelangelo insisted on calling it - were the constant jokes of Raphael, Mikey, and Casey Jones. Michelangelo had a lot of fun with the situation, enjoying every time he used female pronouns to refer to his older brother. Everyone agreed to use "she" and "Leonarda" until Donatello managed to find a solution.
