Peppino is a portly, middle-aged Italian chef who wears a toque, a white tank top over a black t-shirt, black pants and boots. He has black short, messy hair and a small mustache under his long nose. His main emotions are anger and fear, with him often switching between the two in the Pizza Tower.

He owns and works at a restaurant named after himself, titled Peppino's Pizza (occasionally written as Peppino Pizza). The business is in debt and is in a rough state, as shown by the game's introduct

The Vigilante is a yellow Cheeseslime wearing a cowboy hat. He is the grandson of John E. Cheese, the former owner of Fun Farm, which he now owns. He was apparently tricked by Pizzaface into thinking Peppino is a wanted criminal using forgery, leading to his fight at the Western District.

Pepperman is a giant anthropomorphic red bell pepper with limbs wearing white gloves and sneakers of the same color. Pepperman has big, blue eyes and is frequently seen sporting a great, confident grin, showing his teeth. His stem sometimes turns into a beret when he is focused on his artwork or admiring others' work.

His Boss Fight portrait depicts him in a more humanoid appearance, showing him wearing a dark red t-shirt underneath a white apron.

Fake Peppino is an uncanny being that bears a close resemblance to Peppino Spaghetti. He moves disjointedly and seems to be made of a liquid. He is much taller than Peppino with loose eyes and a permanent smile. He also, much like Peppino, wears a white tank top over a black T-shirt, black pants and a white chef's hat; his outfit will change to whatever costume the player is wearing. Additionally, in one of his boss fight attacks, he takes his head off like a mask, revealing nothing pink brain.

Pizzaface is a giant floating cheesy pizza with two red pepperoni eyes, a beige mushroom nose, and a mouth, mustache, and eyebrows made out of green peppers. He can change and move the toppings on his face at will in order to make different facial expressions.

The Noise is a short, mischievous gremlin-like human. He dons a yellow jumpsuit and chef hat-shaped mask, an orange cape, and white gloves. He has a long nose, a thin, curled mustache, and a single bucktooth. The Noise's mannerisms are usually giddy, energetic and child-like. He frequently summons random, often wacky objects out of thin air, as seen in his movesets. Despite his goofy nature and appearance, he can become frighteningly demonic if angered.
