When Kion gave up his position as leader of the Lion Guard to Vitani, his Lion Guard marking on his left shoulder vanished and was replaced with the Night Pride marking on his right shoulder, Kion then gains a blue mark on his forehead symbolizing him as King of the Tree of Life.
