Though badly injured by the near-fatal fall down the cliff, Jill Valentine was kept alive by Albert Wesker, who transported her to Tricell Inc., Africa's top-secret Bio-Organic Weapon (B.O.W.) research outpost near the Kijuju region. She was kept in a medically induced comatose condition while Wesker, inspired by his final encounter with Spencer, began work on the Uroboros Project. This was a catastrophic doomsday plan intended to force a new era of evolution on the survivors using a refined Progenitor strain. Initially, Jill was slated to be the very first test subject for the completed Uroboros, a final, vengeful act by Wesker.

During a preliminary examination, however, a critical discovery was made: the T-Virus vaccination she received back in 1998 had not fully immunized her. A mutant strain of the virus had lain dormant for eight years, only to reactivate while she was in the coma. Jill's highly resilient body quickly fought off the mutant strain, producing a new, extremely pure form of T-Virus antibodies. This anomalous, potent resistance rendered her useless as an early-stage test subject for Uroboros. Recognizing her unique biological value, Wesker spared her life, theorizing that exposure to her antibodies could lead to an Uroboros strain more tolerable to the human body. He kept her alive as a "living factory," repeatedly administering the T-Virus to her blood to continually harvest the antibodies. This constant medical intervention during her stasis caused her hair to turn a distinct blonde and her skin to visibly whiten.

As the Uroboros Project progressed, constant monitoring was no longer required. Jill was released from her captivity, but Wesker had a new purpose for her. He made her the test subject for the P30 Project, a counterpart to Tricell's Plaga Project, which aimed to create super-soldiers with enhanced strength and complete susceptibility to suggestion. The P30 drug, derived from Progenitor research, required continuous administration as it was quickly metabolized. Since there was no data on long-term effects, a specially manufactured device was surgically attached to her chest to periodically inject the drug. Now under Wesker's absolute control, Valentine was informally inducted into the emerging, deadly partnership between Wesker, Excella Gionne, and Ricardo Irving.

Jill is standing beside Wesker as he comes up with his new plans to take over the world.
