Darna is Narda Custodio, a young emergency medical technician, and the successor of Leonor/Zora as the protector of the mythical stone. She is also a master of Kali Martial Arts. She is friends with Brian Robles and Regina Vanguardia. Regina’s alter ego Valentina, is cursed with living, venomous snakes for her hair. Valentina is Darna's archenemy. Despite that information, Narda still loves Regina.

Valentina is a lawyer, philanthropist, social activist, and vlogger who heads the Vanguardia Rescue of Emergency Medical Team Service. She has a secret identity known as Valentina, and she has hallucinations of Valentina taking over her. As Valentina, she is a villain who despises Darna for ruining her plans. She craves to take the white stone from Darna called “ang bato.” Her only powers are launching snakes from her head and making her snakes bite citizens and Darna.

Darna and Valentina are currently fighting a battle against each other but despite that they are archenemies they still love each other deeply.
